Project summary

The purpose of the project is the synthesis Shiff bases of amino acid type and study of their radio protective and antioxidant properties. Synthesized nucleophilic compounds (further on NC) will comprise in their molecules functional groups, contributing to capture of free oxygen forms and binding of water radiolysis products.

In order to characterize obtained NC-s it is planned to study their structure, active centers and other physical-chemical parameters. Synthesized NC-s will be studied as potential radio protectors and antioxidants. In aspect of mentioned properties study will be analyzed their effect for survival, functional activity and by clinic indicators of intact and irradiated animals. On cellular level it is planned to study effect of NC for survival and breeding of intact and irradiated cultivated fiber-blasts. It is proposed to study also influence of NC on sub-cellular level, namely on bio-membranes and chromosomes of cultivated lymphocytes, and on DNA level – by its restrictive specter.

As the result of the project fulfillment will be obtained and characterized new NC-s, will be studied their biologic activity and radiation protective properties. New compositions that have antioxidant and radiation protective properties will be proposed for use in practice.

Project unites the efforts of scientists occupied during different times in the USSR Defense Ministry programs. Project accomplishment will let use the knowledge and experience of researchers, acquired in closed defense laboratories, for pacific medical-biologic tasks solution.

In the project will participate 33 specialists having high education, from which 17 are Candidates of science and 5 are Doctors of science (Ph.D.), and 1 specialist having secondary special education.

This study attributes to basic researches. Data, obtained in the result of research realization, can greatly influence development of activities in the field of organism protection from ionizing radiation. Recommendations will be provided for use of studied agents in medicine and radiobiology.

The project will promote the realization of purposes and tasks of ISTC, as it will provide the possibility for reorientation of scientific personnel activities, created in the frame of defense programs, to fulfill works in the pacific field. The project will help to integrate researchers, that were occupied in weapon activities, into the international scientific community. Proposed research will be of special interest for various fields of organic chemistry and biology, and also for antioxidant and anti-radiation safety means producers.

In the project frame will be solved:

Task 1 – Synthesis and study of physical-chemical properties of ethyl ethers 2-salicyliden tyrosine, tryptophane, histidine, g-aminobutyric acid and their Mg-, Zn-, Co(II)-, Fe(II)-salts.

Study of medical-biologic and radio protective characteristics of the synthesized NC-s in intact and irradiated animals (NC toxicity, their influence on survivability and general clinic-physiologic parameters of animals).

Study of biologic activity of radio protective and antioxidant properties of NC for cellular and non-cellular experimental models (cultures of vertebrates’ cells, bio-membranes, DNA).

Task 2 – Synthesis and study of physical-chemical properties of ethyl ethers 2-pyridiliden tyrosine, tryptophane, histidine, g-aminobutyric acid and their Mg-, Zn-, Co(II)-, Fe(II)- salts.

Study of medical-biologic and radio protective characteristics of the synthesized NC-s in intact and irradiated animals (NC toxicity, their influence on survivability and general clinic-physiologic parameters of animals).

Study of biologic activity of radiation protective and antioxidant properties of NC for cellular and non-cellular experimental models (cultures of vertebrates’ cells, bio-membranes, DNA).

For the solution of the stated tasks it will be used the complex methodical approach.

The following methods will be used: organic synthesis, study of structure and physical-chemical properties of NC-s (NMR, EPR, spectroscopy), analysis of survivability, functional activity and clinic indicators of animals, determination of survivability and rate of breeding of cultivated cells, analysis of structural and functional characteristics of bio-membranes, study of chromosomes and DNA.

The foreign collaborators will take part in data exchange during the project conduct, comments presentation in addition to project participants reports and conduct of joint work seminars.

The scope of the work is estimated for 552 man-months.
