Project summary

The project objective is to reveal mechanisms of the damaging action of physical and chemical factors (gamma, UV and laser radiation, phenylhydrazine and tetraethylstannum) on the nervous, immune and haemopoietic systems of mammals and of the protective influence of biologically active compounds (as synthesized by the project team), condensed thienepyrimidines, and pyracetam, diazepam, Ca-ds-RNA and sodium thiosulphate. This work can be split into the solution of three tasks, as follows:

1. Development of convenient and effective methods of synthesis of substituted thiene (2,3-d) pyrimidines condensed with hexatomic heterocycles, capable of rendering a protective influence on the functioning of the nervous, immune and haemopoietic systems.
2. Detection and research of disturbances in cells of the nervous, immune and hemopoietic systems and liver under the action of the tested physical and chemical damaging factors.
3. Determination of the efficiency of application of biologically active compounds (protectors) with the purposes of weakening (preventing) the damaging action of extreme factors and reducing the normal functioning of the nervous, immune and hemopoietic systems.

The topicality of the problem is defined by the increasing danger of radiation, laser and chemical influence and the need for the development of new methods of organism protection and of recovery of the normal functioning of systems after damage. This primarily concerns the influence of laser radiation, where a full range of questions, connected with differentiated determination of the damaging factor in continuous and impulse modes, has been practically not studied at all. The influence of toxic compounds of industrial and usual-life origin, could also bring serious consequences, as could neuropathic toxins, the production and storage of which could be connected with dangerous situations. This explains the selection of damaging factors and of model systems with acute reaction on their influence.

Scientific validity of project
The project plans fundamental studies, by which unstudied or little studied molecular-cellular mechanisms of cell and cell population behavior regulation, under the influence of damaging physical-chemical factors in sub lethal doses, will be revealed. In the first instance this is related to the virtually unstudied influence of laser radiation. At the same time, data will be obtained on the influence in these conditions of biologically active substances on the regulation mechanisms of cellular activity. In the first instance, the influence of condensed heterocyclic compounds with protective features (synthesized by the authors for the first time), and their role in the restoration of the normal functioning of the nervous, immune and haemopoietic systems, is to be studied. On the basis of these studies, performed by qualified experts with the use of modern analytical techniques, the methodology of complex protection and restoration of organisms of mammals from the influence of damaging factors will be developed.

Practical significance of the project is defined by creation of:
а) new-generation condensed heterocyclic systems with protective features;
b) complex of drugs with combined protective and cell structure damage reducing properties;
c) methodo-logy for tissue-specific protection and reduction of damaged cells and their populations on the basis of stimulation of the activity of endogenous protective mechanisms with the help of the exogenous influence of biologically active substances, developed and tested by the project team.

Commercialization of results
A large number of condensed heterocyclic compounds, synthesized in the course of project performance, and after analysis of their correcting, stimulating and protective properties, will be recommended for testing as pharmaceuticals and can be demanded by interested medical and commercial organizations.
The project tasks include the reorientation of scientists, previously employed in defense programs, and the creation of a strong scientific collective of highly qualified professionals, for versatile research in common areas of physics, chemistry and biology, to define the degree of influence of extreme factors on both cell and molecular frames of mammals and the development of means to overcome the consequences of damage and reduction of their functioning.
