Project summary

The searching of cells, sensitive to viruses is one of the basic problems in virology and biotechnology. It has become extremely urgent in the last decade in connection with the appearance of new virus infections (AIDS, hepatitis C, D, E, G, etc.), pathogen for the human. This problem has great importance, as it is directly connected to the manufacture and control of immunobiological preparations (production of a virus antigen for the diagnostic test systems and the manufacture of live vaccines). Nowadays, for selection of isolates of a virus a very enduring and laborious method related with heuristic search of sensitive to a virus cells is used.

In SRC VB «Vector» experimental data about significant and fast changes of electrophyisical characteristics (polarization degree) of cells infected by a virus are gotten. These changes are observed already at the stage of viruses adsorption to the cell membrane and connected with considerable distinction in behavior of intact and infected cell in non-uniform alternative electric field (NUAEF).

The project goals:


1. To develop an express method of identification of cells infected by a virus with the help of a non-uniform alternative electrical field;
2. To develop a method of determining of concentration of a virus in a cell suspension with the help of a non-uniform alternative electrical field.

These methods will find wide application both in scientific research, and in manufacture of vaccines. They will allow searching the cells sensitive to viruses in a very short period of time.

In performance of this project, 32 specialists will be involved who were previously working in the field of biological protection. This group has more than ten years experience in theoretical and experimental research with virus cell interaction. The realization of the project will allow to reorient their abilities for development of economical tasks, that meet the requirements ISTC.

The methodology of researches is based on measurement of value of electrical polarization (induce dipole moment) and study of behavior of intact and infected cell in non-uniform alternative electrical field. Herewith the quantity observable under a microscope depolarized (infected) cells relates to concentration of a virus in cell suspension. It is supposed to carry out measurement of dipole moment and research of behavior of a separate cell with the help of laboratory device, which will be consist of: generators of a variable voltage, microscope with a videocamera, monitor, videorecorder, computer, and also measuring device. The calculation of number of intact and infected cells is supposed to be automated by use specialized microscope CMS-700 from «Scion Corporation» and specialized computer program.

The participation of Collaborator is supposed in the project:


– in discussion of the received results;
– in independent check of the received experimental data;
– in information exchange;
– in representation of the comments to the technical reports (quarter, annual, final etc.);
– in realization of joint symposiums and working seminars.
